Micronuclei formation in liver fibrosis samples from patients infected by hepatitis C virus Genet. Mol. Biol.

Genetic research on fibrosis outset and its progression in chronic hepatitis (CH) by hepatitis C virus (HCV) are limited. The lack of cytogenetic data led us to investigate the presence of micronuclei (MNi), as a sign of genomic damage. Hepatocytes of hepatic parenchyma from 62 cases diagnosed with CH associated with HCV and displaying different degrees of fibrosis (F1-F4) were analyzed. These data were compared to 15 cases without fibrosis (F0). Twelve healthy liver parenchyma samples were included as control. All samples were obtained from paraffin-embedded archival material. Adaptation or acclimation to hypoxia occurs via the modulation of physiologically relevant genes, such as erythropoietin, transferrin, vascular endothelial growth factor, phosphofructokinase and lactate dehydrogenase A. In the present study, we have cloned, sequenced and examined the modulation of the LDH-A gene after an Amazonian fish species, Astronotus crassipinis (the Oscar), was exposed to hypoxia and anoxia. In earlier studies, we have discovered that adults of this species are extremely tolerant to hypoxia and anoxia, while the juveniles are less tolerant. Identificação de genótipos de feijão-caupi tolerantes a salinidade avaliado por meio de método multivariado Ciência Rural

Objetivou-se com este trabalho identificar genótipos tolerantes ao estresse provocado pela solução de NaCl e pela mistura de NaCl, CaCl2 e MgCl2 na água de irrigação, nos estádios iniciais de desenvolvimento. Os níveis de condutividade elétrica do extrato de saturação foram: 0, 2,5; 5,0 e 7,5dS m-1. A tolerância do feijão-caupi foi avaliada através da redução relativa de matéria seca da parte aérea de cada genótipo. O genótipo CE-182 mostrou-se mais tolerante nos níveis 2,5; 5,0 e 7,5dS m-1 na água salinizada com solução de NaCl. First record of Wolbachia in South American terrestrial isopods: prevalence and diversity in two species of Balloniscus (Crustacea, Oniscidea) Genet. Mol. Biol.

Wolbachia are endosymbiotic bacteria that commonly infect arthropods, inducing certain phenotypes in their hosts. So far, no endemic South American species of terrestrial isopods have been investigated for Wolbachia infection. In this work, populations from two species of Balloniscus (B. sellowii and B. glaber) were studied through a diagnostic PCR assay. Fifteen new Wolbachia 16S rDNA sequences were detected. Wolbachia found in both species were generally specific to one population, and five populations hosted two different Wolbachia 16S rDNA sequences. Genetic structure and demographic history of striped weakfish Cynoscion guatucupa (sciaenidae) from the southwestern atlantic JBAG

In South America, the Pleistocene was characterized by important environmental changes related to glacial cycles, which had significant effects on the evolutionary history of several marine species. To evaluate the pattern of demographic history and the genetic structure of striped weakfish Cynoscion guatucupa, a 401 bp fragment of the mitochondrial Cytochrome b gene was sequenced from 92 individuals from three coastal areas in Argentina and one in Brazil in the southwestern Atlantic. The amplified fragment length polymorphism (AFLP) assay is an efficient method for the identification of molecular markers, useful in the improvement of numerous crop species. Bulked Segregant Analysis (BSA) was used to identify AFLP markers associated with water-stress tolerance in barley, as this would permit rapid selection of water-stress tolerant genotypes in breeding programs. Chromosome identification in the Andean common bean accession G19833 (Phaseolus vulgaris L., Fabaceae) Genet. Mol. Biol.

Characterization of all chromosomes of the Andean G19833 bean genotype was carried out by fluorescent in situ hybridization. Eleven single-copy genomic sequences, one for each chromosome, two BACs containing subtelomeric and pericentromeric repeats and the 5S and 45S ribosomal DNA (rDNA) were used as probes. Comparison to the Mesoamerican accession BAT93 showed little divergence, except for additional 45S rDNA sites in four chromosome pairs. Altogether, the results indicated a relative karyotypic stability during the evolution of the Andean and Mesoamerican gene pools of P. vulgaris.

Sequences potentially associated with coffee resistance to diseases were identified by in silico analyses using the database of the Brazilian Coffee Genome Project (BCGP). Keywords corresponding to plant resistance mechanisms to pathogens identified in the literature were used as baits for data mining. Uno de los materiales de papa más susceptibles a los virus corresponde a Solanum phureja; sin embargo, en Colombia son pocos los estudios adelantados sobre los agentes causales que lo afectan. Microsatellite polymorphism was studied in a sample of 39 traditional rice (Oryza sativa L.) varieties and 11 improved varieties widely planted in Cuba. The study was aimed at assessing the extent of genetic variation in traditional and improved varieties and to establish their genetic relationship for breeding purposes. Heterozygosity was analyzed at each microsatellite loci and for each genotype using 10 microsatellite primer pairs. Between varieties genetic relationship was estimated. The number of alleles per microsatellite loci was 4 to 8, averaging 6.6 alleles per locus. Comparative karyotype analysis in diploid and triploid Dolichoplana carvalhoi (Tricladida, Terricola, Rhynchodemidae) from Brazil Genet. Mol. Biol.

In this work, we present cytogenetic data for the land planarian Dolichoplana carvalhoi. Two different karyotypes, one diploid (2n = 2x = 14 chromosomes) and one triploid (2n = 3x = 21 chromosomes), corresponding to two morphological body patterns, are described. Chromosomes from regenerating blastema were studied after routine Giemsa staining and CBG banding. Our analyses revealed heteromorphisms in chromosomes 2, 3 and 4 of the diploid karyotype and in chromosomes 1, 3 and 7 of the triploid karyotype. Further studies are needed in order to determine if the two morphological patterns of D. carvalhoi represent distinct species.

There is considerable controversy with regard to the genotoxicity of glyphosate, with some reports stating that this compound is non-toxic for fish, birds and mammals. In this work, we used the comet assay to examine the genotoxicity of glyphosate isopropylamine (0.7, 7, 70 and 700 µM) in human lymphocytes, erythrocytes of Oreochromis niloticus and staminal nuclei of Tradescantia (4430) in vitro and in vivo. Cells, nuclei and fish that had and had not been exposed to 5 mM N-nitrosodiethylamine (NDEA) were used as positive and negative controls, respectively. Glyphosate is noted for being non-toxic in fishes, birds and mammals (including humans). Nevertheless, the degree of genotoxicity is seriously controversial. In this work, various concentrations of a glyphosate isopropylamine salt were tested using two methods of genotoxicity assaying, viz., the pink mutation assay with Tradescantia (4430) and the comet assay with nuclei from staminal cells of the same plant. Staminal nuclei were studied in two different forms, namely nuclei from exposed plants, and nuclei exposed directly.
